چکیده

Human apolipoprotein A-IV is an acidic polypeptide of molecular weight 46,000 that is secreted into lymph on the surface of nascent chylomicrons, but which exists in circulation unassociated with lipoproteins. Previous studies of this protein have utilized material isolated from a d < 1.006 g/ml fraction of human serum and from human lymph. Although it has been suggested that apoA-IV circulating in plasma is the product of dissociation from the surface of nascent chylomicrons, it has been characterized by immunological techniques only. We have isolated human apoA-IV on a preparative scale from lipoprotein-depleted serum using a technique of adsorption to a phospholipid-triglyceride emulsion, followed by delipidation and preparative gel electrophoresis. The molecular weight, pI, and amino acid composition of material thus prepared agree with literature values for apoA-IV derived from chylomicrons. We have determined that apoA-IV is a glycoprotein containing 6% carbohydrate by weight (mannose 1.8%, galactose 1.55%, N-acetyl glucosamine 1.55%, sialic acid 1.1%). Electroimmunoassay of human serum using a monospecific rabbit antibody to serum-derived apoA-IV found 13.1 +/- 1.8 mg/dl, a value in agreement with determinations using antibodies to chylomicron-derived apoA-IV. We conclude that apoA-IV may be easily purified from normal human serum, and that the material thus isolated has identical chemical, physical, and immunological properties to apoA-IV obtained from human lymph. It is therefore likely that the dissociation of apoA-IV from the surface of nascent chylomicrons following their entry into circulation is not attended by changes in the structure or composition of this apoprotein.-Weinberg, R. Evidence for high density lipoproteins as the major apolipoprotein A-IV-containing fraction in normal human serum.

The distribution of human apolipoprotein A-IV was studied in sera from normolipidemic fasting subjects by high performance gel filtration on a Superose 12 HR column. The major part of apolipoprotein A-IV eluted in the range of the apolipoprotein A-I peak, and distributed mainly in the large-size high density lipoprotein subfractions. Only a small peak or a shoulder on the main fraction appeared...

Identification of Sequence Variation in the Apolipoprotein A2 Gene and Their Relationship with Serum High-Density Lipoprotein Cholesterol Levels

Background: Apolipoprotein A2 (APOA2) is the second major apolipoprotein of the high-density lipoprotein cholesterol (HDL-C). The study aim was to identify APOA2 gene variation in individuals within two extreme tails of HDL-C levels and its relationship with HDL-C level. Methods: This cross-sectional survey was conducted on participants from Tehran Glucose and Lipid Study (TLGS) at Research Ins...

Importance of evaluating cell cholesterol influx with efflux in determining the impact of human serum on cholesterol metabolism and atherosclerosis.

OBJECTIVE Cholesterol efflux relates to cardiovascular disease but cannot predict cellular cholesterol mass changes. We asked whether influx and net flux assays provide additional insights. APPROACH AND RESULTS Adapt a bidirectional flux assay to cells where efflux has clinical correlates and examine the association of influx, efflux, and net flux to serum triglycerides (TGs). Apolipoprotein ...

Apolipoprotein E Polymorphism in an Iranian Hypercholestrolemic Population

Apolipoprotein E (apo E) is a structural constituent of several serum lipoprotein classes. It plays an important role in lipid metabolism by acting as a ligand for low-density lipoprotein (LDL) and chylomicron remnant receptors. Three common alleles called e2, e3 and e4 have been described, which code for three protein isoforms (E2, E3 and E4). The polymorphism is clinically significant, and it...
